The phase I and II trials focus on combining drugs to slow the development of resistance to drugs that inhibit BRAF, a gene that is mutated in about half of melanomas. Earlier trials with drugs that target BRAF generated excitement for their ability to quickly shrink melanoma tumors in suitable patients. But for many patients the benefits proved short-lived, as the cancer cells develop resistance to the drugs.
"These studies exemplify an important landmark of some tumors, which has emerged from recent laboratory research: the presence of specific mutations, such as the BRAF mutation in metastatic melanoma which exposes an Achilles' heel--MEK in this case," said Prof Yossef Yarden from the Weizmann Institute of Science, Israel. "In-depth understanding of cancers and their mutations is expected to reveal more of these deadly weaknesses in cancer, which we can exploit using new drugs and drug combinations."
Phase II of the BRAF inhibitor dabrafenib alone vs combination with MEK1/2 inhibitor trametinib
Dr Georgina Long from Westmead Hospital and the Melanoma Institute Australia and colleagues report that combining the new drugs dabrafenib and trametinib provided a clinically meaningful improvement in progression-free survival, response rate and duration of response in 162 patients with melanoma that had BRAF V600 mutations.
Patients in the study received either dabrafenib 150mg twice daily; twice-daily dabrafenib plus once-daily 1mg trametinib; or twice daily dabrafenib plus once-daily 2mg trametinib. The combination prolonged progresion free survival over single-drug therapy from 5.8 months to 9.4 months, which represents a 60% improvement. Among patients who received both drugs at the higher dose, 41% had not progressed 12 months after treatment began, compared to 9% in the monotherapy arm of the study.
"The combination therapy of the BRAF inhibitor dabrafenib and the MEK inhibitor trametinib prolongs the progression-free survival in patients with V600 BRAF mutation-positive metastatic melanoma compared with dabrafenic monotherapy," Dr Long said. "Importantly, the combination also decreases the rate of the cutaneous toxicities compared with dabrafenib monotherapy, particularly the oncogenic cutaneous toxicity of squamous cell carcinoma."

A Phase I study in 44 patients shows that the combination of the MEK inhibitor GDC-0973 and vemurafenib can be delivered safely, Dr Rene Gonzalez of the University of Colorado Cancer Center, Denver, and colleagues report.
"BRAF inhibition has resulted in high response rates and improved survival in patients with BRAF mutated melanoma," Dr Gonzalez said. "One of several mechanisms of resistance has been reactivation of the MAPK pathway. Preclinical models show that combined inhibition of BRAF and MEK can delay the acqusition of resistance compared to BRAF inhibitor monotherapy. Inhibition of the pathway downstream from BRAF with the MEK inhibitor GDC-0973 could theoretically overcome or delay this resistance mechanism and improve outcomes."
The study was not designed to evalate efficacy. "While early data in a small number of patients did show tumor reduction, it would be premature to comment on efficacy based on these preliminary results and further research is warranted," Dr Gonzalez said.

LOS ANGELES (Reuters) - Drivers in Los Angeles saw limited congestion on Saturday near a key freeway corridor shut down this weekend in what has been dubbed "Carmageddon 2," due to fears of gridlock from a 53-hour construction project, officials said.
A 10-mile (16-km) stretch of the 405 Freeway at the Sepulveda Pass, normally the nation's busiest thoroughfare, was closed at midnight on Friday for a $1 billion freeway widening project.
But so far, despite the slowing near the project itself, traffic seemed to be zipping along in the city as a whole, said California Highway Patrol spokeswoman Officer Charmaine Fajardo.
"So far so good, it looks like people are heeding the information we've put out there and staying out of the area," Fajardo said.
Construction crews with giant cranes on Saturday chipped away at a bridge spanning the 405 Freeway in the city, with pieces of concrete dropping dozens of feet (meters) to the ground below. A four-foot (1.2-metre) pad of dirt laid down in advance protected the pavement from the falling debris.
The work is to widen the bridge itself to expand the freeway below.
The project comes just over a year after a similar weekend construction shut down the same area in what was dubbed "Carmageddon," in anticipation of massive congestion that never came to pass as many drivers stayed home.
This year, the project has caused some anxiety nonetheless among officials and motorists in the city where car travel is king but only very limited congestion had materialized by noon.
More significant delays could develop on Saturday afternoon as people pile into cars heading for the beach, highway patrol officials said.
The shutdown of the 405 Freeway along the Sepulveda Pass is scheduled to end at 5 a.m. on Monday, said Marc Littman, a spokesman for the Metropolitan Transportation Authority.
The construction crew on the project stands to be fined $6,000 per closed lane for every 10 minutes that the project extends past its scheduled ending on Monday morning, officials said. (Reporting by Alex Dobuzinskis; Editing by Colleen Jenkins and Sandra Maler)

BAGHDAD (AP) ? Iraqi officials said Saturday that a jailbreak where al-Qaida-linked militants escaped death row had help from inside, further tarnishing state authority and raising new concerns over corruption.
A day after the escape in Tikrit, Saddam Hussein's hometown 130 kilometers (80 miles) north of Baghdad, scores of prisoners are still at large.
The Interior Ministry said there had been "clear collusion" between some guards and inmates in the Tasfirat prison. Weapons were brought in during family visits, and wardens left locks inside the facility open.
"The cells were not searched for a long period, which indicates more deliberate negligence that led to this incident," the ministry said in a statement released late Friday.
The escape occurred after a riot and firefight that left 20 dead, including 16 inmates and four guards, the statement said. After taking over a large part of the prison, rioters used other inmates as human shields in order to make their way out, it added. Of a total 303 prisoners, 102 escaped, including 47 al-Qaida-linked inmates awaiting execution. Some 23 were recaptured.
On Saturday, state television announced a reward for information leading to the arrest of the fugitives, and a curfew remained in force until the afternoon. Salahuddin provincial spokesman Mohammed al-Assi said "security forces have intensified efforts to hunt down those still on the run."
He refused to say how much the reward is for.
The incident generated sharp criticism for Iraqi security forces, who have been unable to stabilize the country almost a year after the withdrawal of U.S. troops. Several smaller escapes and attempted jailbreaks have already deeply embarrassed the government, which is eager to demonstrate it can run its justice and detention systems.
The Interior Ministry statement also said that Maj. Gen. Abdul-Karim al-Khazraji, the police chief of Salahuddin province where the jailbreak occurred, was fired.
Iraq has a poor track record of keeping inmates behind bars.
The Tikrit prison itself was moved to a different location after 16 prisoners, including five al-Qaida-linked inmates awaiting execution, made their escape after prying open the bars on a prison bathroom window with a pipe wrench in September 2009.
At the time, the entire prison staff and the provincial prison official were detained for questioning. Six of the escaped inmates were later captured.
And in southern city of Basra, a dozen detainees held on terrorism charges broke out of a prison in 2010, disguised in police uniforms. And last year, al-Qaida smuggled weapons and grenades into a prison in Mosul, supposedly one of the country's most secure detention centers, and attempted an ultimately unsuccessful jailbreak that left 17 dead.
In July 2011, detainees linked to al-Qaida escaped at least twice from a Baghdad area prison known as Camp Cropper shortly after the U.S. handed it over to Iraqi authorities.
Authorities earlier reported 12 people dead and 32 wounded in Friday's break in Tikrit.

The United States sent Guant?namo?s youngest captive home to a prison in his native Canada on Saturday, ending the decade-long U.S. detention of the Muslim militant who grew from a teenager into adulthood at the Pentagon?s prison camps in Cuba.
In 2002, at age 15, Omar Khadr hurled a grenade in war-torn Afghanistan that killed an American soldier. He was captured and kept by U.S. forces ever since. Now 26, he?s home and could serve up to six more years in a Canadian prison. Or authorities could release him sooner. Canada considers him a juvenile offender who can apply for parole a year from now.
?He thinks he?s in a dream. He?s pinching himself,? said Toronto attorney John Norris, who spoke to Khadr soon after his arrival. ?He never believed this day would come; he?s been betrayed so many times before.?
Khadr departed the base in Cuba before dawn Saturday, a secret transfer 10 days after a Canadian diplomat paid Khadr a visit on his 26th birthday. He landed at a Royal Canadian airbase in Ontario and was transferred to the Millhaven maximum security prison for what his lawyer described as an assessment of the most suitable place to serve out his sentence.
The case of Khadr ? Guant?namo?s last Western captive ? stirred debate in international law and human rights circles.
Because he was captured at such a young age, some called him a child soldier who was dropped off in the war zone by his father and deserving of rehabilitation not interrogation. Others called him the respected scion of an al Qaida family, nicknamed Canada?s First Family of Terror in news reports, and opposed his repatriation.
Psychiatrist Michael Welner, testifying at the Guant?namo war court for the prosecution and paid by the Pentagon, called Khadr a continuing danger who spent his time at the U.S. prison camps in Cuba ?marinating in a community of hardened and belligerent radical Islamists.?
Khadr?s lawyers offered up tales of abusive treatment in U.S. custody. He?d been questioned in Afghanistan while emerging from surgical anesthesia, they argued. He was used as a ?human mop? at Guant?namo after he urinated on himself and the floor during an interrogation. His lawyer was forbidden from seeing him during his first two years of interrogations.
U.S. troops captured Khadr, who was near death, in a July 27, 2002 firefight at a suspected al Qaida compound near Khost, Afghanistan.
U.S. air strikes had leveled the compound and as a Special Forces unit assaulted, Khadr admitted in a 2010 guilty plea, he threw a grenade from the rubble that mortally wounded Army Sgt. 1st Class Christopher Speer, 28. Medics were able to save Khadr?s life and turned him over to what became a decade of on-again off-again interrogation.
Since his conviction, he was confined to a maximum-security wing at Guant?namo, a mostly solitary existence that his lawyers tried to ameliorate with a Canadian-style college preparatory curriculum ? literature, physics and videos of ?Little Mosque on the Prairie,? a popular Canadian TV show about a Muslim community in a fictional prairie town.
He also read Shakespeare with his U.S. defense lawyer, Army Lt. Col. Jon Jackson, who played Juliet to Khadr?s Romeo.

WASHINGTON (Reuters) - A Virginia auction house on Thursday canceled the sale of a Renoir painting bought at a flea market for $7 after signs the work was stolen from the Baltimore Museum of Art decades ago.
The painting "Paysage Bords de Seine" by Pierre-Auguste Renoir was to have gone under the hammer on Saturday but ownership questions halted the sale, said Lucie Holland, a spokeswoman for Potomack Co, the Alexandria, Virginia, auctioneer.
"The rest of the auction will go on, but the Renoir has been withdrawn," she said.
A Virginia woman bought the signed French Impressionist painting at a West Virginia flea market a year or two ago, hoping the frame would be of some use.
She ignored the work until it turned up again while housecleaning and had it appraised by Potomack in July. The auctioneers verified it as a Renoir and estimated its worth at $75,000 to $100,000.
The Baltimore Museum of Art told Potomack on Wednesday that its records indicated "Paysage Bords de Seine," or "Landscape on the Banks of the Seine," was stolen while on loan to the museum in 1951, the auction house said.
Potomack told the FBI and a federal probe is under way. There is no known police report on the theft.
BOUGHT IN PARIS
The Renoir came to the Baltimore museum through one of its leading benefactors, collector Saidie May. Her family bought the painting from the Bernheim-Jeune gallery in Paris in 1926.
The Washington Post found records in the museum's library on Tuesday that showed May had lent the paintings and other works to the museum in 1937, Potomack said.
After the newspaper told it of the findings, the Baltimore museum checked its files and found a loan record showing the Renoir had been stolen on November 17, 1951. What happened to it after the theft is unknown.
Doreen Bolger, the museum director, said the museum's probe into what happened to the painting was in early stages but was centered on May.
She died in May 1951 and the art collection was willed to the museum. As its ownership was going through legal transfer, the painting was stolen while still listed as on loan.
"At this point we just want to make sure that the painting winds up where it belongs and that we provide all the information we can to law enforcement about this issue," Bolger said.
Potomack said the painting had not turned up when it checked London's Art Loss Register, a database of stolen and lost art. It also consulted the FBI's art theft website to confirm it was not listed as stolen.

I would like to take this blog entry to inform you of some important announcements for further renovations and maintenance around the Sports and Recreation facilities.
The 3/F Swimming Pool will be closed from October 8 to 24.? As well as the installation of the bubble, we will carry out essential maintenance. During this period the G/F Tropical Pool will be open from 7am to 10.30pm daily.
North Shore Tennis Court will be renovated from mid November to mid January and one court will be kept open for Members.
Prior to the closure of the 3/F Swimming Pool as mentioned above I would like to remind you that our Annual Swim Gala will take place on Sunday, October 7 from 1pm to 4pm.
The Bowling Alley renovation is scheduled to be ready towards the latter part of October and we are also looking forward to introducing a new Table Tennis room on the 7/F in the coming weeks.
Please also look out for our new group exercise classes that we will be introducing in the near future such as Fitball and ViPR (Vitality, Performance and Reconditioning).

"Today we're looking at science fiction becoming tomorrow's reality ? the self-driving?car," Brown said. "Anyone who gets inside a?car?and finds out the?car?is driving will be a little skittish, but they'll get over it."
Google has been developing?autonomous?car?technology and lobbying for the regulations. The company's fleet of a dozen computer-controlled vehicles has logged more than 300,000 miles (483,000 kilometers) of self-driving without an accident, according to Google.
"I think the self-driving?car?can really dramatically improve the quality of life for everyone," Google co-founder Sergey Brin said.
Autonomous?cars?can make roads safer, free commuters from the drudgery of driving, reduce congestion and provide transport to people who can't drive themselves, such as the blind, disabled, elderly and intoxicated, Brin said.
"I expect that self-driving?cars?will be far safer than human-driven?cars," Brin said.
Brin predicted that?autonomous?vehicles will be commercially available within a decade. He said Google has no plans to produce its own?cars, but instead plans to partner with the automobile industry to developautonomous?vehicles.
The Alliance of Automobile Manufacturers expressed concern that California is moving too quickly to embrace self-driving?cars?and needs to first sort out liability issues.
"Unfortunately this legislation lacks any provision protecting an automaker whose?car?is converted to anautonomous?operation vehicle without the consent or even knowledge of that auto manufacturer," the trade group said in a statement.
Autonomous?cars?use computers, sensors and other technology to operate independently, but a human driver can override the autopilot function and take control of the vehicle at any time.
With smartphone-wielding drivers more distracted than ever, backers say robotic vehicles have the potential to significantly reduce collisions and traffic fatalities, noting that nearly all?car?accidents are a result of human error.
The legislation requires the California Department of Motor Vehicles to draft regulations for?autonomousvehicles by Jan. 1, 2015. Currently, state law doesn't mention self-driving?cars?because the technology is so new.
The regulations would allow vehicles to operate?autonomously, but a licensed driver would still need to sit behind the wheel to serve as a backup operator in case of emergency.
The legislation also is aimed at keeping California at the forefront of the?autonomous?car?industry since Stanford University and Silicon Valley companies have been working on the technology for years.
In February, Nevada became the first U.S. state to approve regulations spelling out requirements for companies to test driverless?cars?on that state's roads.
Carmakers such as Audi AG, BMW AG, Ford Motor Co. and Volvo have been working on?autonomous?cartechnology for years.
In recent years, automakers also have been introducing?autonomous?functions such as self-parking, lane departure warnings and adaptive cruise-control, which allows vehicles to automatically accelerate and decelerate with the flow of traffic.
Outside a cafe in Mountain View, California, customers said they looked forward to a day when their?cars?could drive themselves, as long as they could do it safely.
"It would make our streets safer," said Barrett Howard, 33, an auto technician. "We wouldn't have to worry about people texting or getting sidetracked. The computer will take over, and it will make life easier."
Abraham Eshel, a retired mechanical engineer who has vision problems, said self-driving?cars?would make his life easier, too.
"If I could tell my?car, 'OK, you take over when it's dark,' and I don't have to worry about it, that would be fantastic," said Eshel, who lives in nearby Los Altos. "It's a good idea. Why not make progress if it's possible?"

We wanted to (not?) see that for ourselves at the Paris Auto Show, so took a quick tour of the magic LED cloak and the technology behind it. It doesn't work quite as well in a show hall as it did when we first saw it in its natural habitat, but the system was still a fun way to show off Merc's green ambitions. It works by passing video from behind the car taken with a Sony video camera through a laptop to a 200 x 300 resolution LED curtain. That makes the car blend in with its background, which is what such a car would do in the real world as far as its emissions go -- apart from a little water, of course. MADRID (AP) ? Spain's Parliament took on the appearance of a heavily guarded fortress Tuesday, hours ahead of a protest against the conservative government's handling of the economic crisis.
The demonstration, organized behind the slogan 'Occupy Congress,' is expected to draw thousands of people from around Spain and was due to start around 1730 GMT.
Madrid's regional Interior Ministry delegation said some 1,300 police would be deployed though protesters say they have no intention of storming the chamber, only of marching around it.
They are calling for fresh elections, claiming the government's austerity measures show the ruling Popular Party misled voters to get elected last November.
The protest comes as Spain struggles in its second recession in three years and with unemployment near 25 percent.
Spain has introduced austerity measures and economic reforms in a bid to convince its euro partners and investors that it is serious about reducing its bloated deficit to 6.3 percent of gross domestic product in 2012 and 4.5 percent next year.
Concerns over the country's public finances was evident earlier when the Treasury sold ?3.98 billion ($5.14 billion) in short-term debt but at a higher cost.
It sold ?1.39 billion in three-month bills at an average interest rate of 1.2 percent, up from 0.95 percent in the last such auction Aug. 28, and ?2.58 billion in six-month bills on a yield of 2.21 percent, up from 2.03 percent.
The government is expected to present a new batch of reforms Thursday as it unveils a draft budget for 2013. A day later the results of bank stress tests carried out by an international auditing company are to be released.
Spain has already been granted a ?100 billion loan by its 16 partner nations using the euro currency to help bail out those of its banks worst hit by the collapse of the country's real estate sector in 2008.
Spain has yet to tap the fund and initial estimates say the banks will need some ?60 billion.

For decades, breast cancer was considered a single disease. Recent research, however, suggested it was a number of genetically distinct variations?and now a new study has split the disease into four differently treatable types.
The study, published yesterday in Nature, is the first comprehensive genetic analysis of breast cancer to date. It suggests that four distinct types of the cancer display hallmark genetic changes, that in the future will be targeted by different treatments.
Part of a project called the Cancer Genome Atlas, designed to build comprehensive maps of genetic changes in common cancers, the study included analysis of tumors from 825 patients. While the researchers found at least 40 genetic alterations that might be attacked by drugs, four of the genetic effects are fundamentally different between the four identified cancer types. Joe Gray, a genetic expert at Oregon Health & Science University, told the New York Times:
"We now have a good view of what goes wrong in breast cancer. We haven't had that before."
Amazingly, the finding offers some immediate implications. The results suggest that routine treatments often used in ovarian cancer could instead be applied to some breast cancers. As ever, though, it's best not to get too over-excited by the finding: while ground-breaking, the finding acts as more of a starting point.
Indeed, the researchers are keen to point out that it will take years to translate some of the new findings into safe, targeted treatments, especially given that individual cancerous tumors of the same type can vary so much case-to-case. But in the longer term, the finding opens up the possibility of diagnosis made on the back of genetic analysis, and tailored treatments plans of drugs and other therapies. The sooner, the better. A Turkish court heard final arguments Thursday in the trial of hundreds of active and retired military officers accused of plotting to overthrow the Islamic-rooted government.
The two-year-long case is wrapping up at the court in Silivri, near Istanbul, which heard the final testimonies of the suspects in the so-called "Sledgehammer" trial, named after a 2003 military exercise.
Prosecutors have demanded up to 20 years in prison for the 365 military officers in the case, which concerns alleged army plans to bomb historic mosques in Istanbul and spark conflict with neighbouring Greece to facilitate a military coup.
The defendants argue that the alleged plot was a military exercise regularly held by the army, and question the authenticity of some documents presented as evidence.
A final verdict was expected later Thursday, the first from a series of cases targeting the army.
The trial, which began in December 2010, stands out in Turkish politics because it directly attacks the secular army, once untouchable in the predominantly Muslim country, where it authored four coups in half a century.
At Thursday's hearing, the judge listened to the defence of Cetin Dogan, former commander of the First Army and suspected of being the "mastermind" behind the 2003 plan to drive the ruling Justice and Development Party (AKP) from power.
"Here we see a process unfolding to make the soldiers of Mustafa Kemal (Ataturk, the founder of modern Turkey), who give their lives for their country, to pay the price of their commitment to the Republic and its principles," said the ex-general, according to the written transcript of his defence.
Dogan branded the trial as "unfair and unlawful," claiming it had been launched by supporters with "a mentality considering all those who do not belong to their brotherhood as enemies."
His remarks will likely be seen as an implicit reference to the AKP government and the influence on the judiciary of Islamic cleric Fethullah Gulen's religious movement.
Around 250 of the defendants are standing trial under arrest.
All the defendants have denied the charges and branded evidence as fabricated. Their lawyers were absent from the courtroom in a protest against what they dubbed a lack of willingness on the part of the court to verify the authenticity of documents presented.
"The verdict given will not be lawful but political. We are the victims of a settlement with the army," said General Bilgin Balanli.
Hundreds of suspects, including army officers, journalists and lawmakers, are being tried separately over their alleged role in plotting to topple the Islamic-rooted government.
Pro-government circles have praised the trial as a step toward democracy, but pro-secular ones have branded it a witch-hunt to silence opposition.
The coup trials are also seen as part of an effort by the current AKP government to roll back the military's influence in politics.
The Turkish army, which sees itself as the guarantor of Turkey's secular principles, overthrew three governments in 1960, 1971 and 1980.
And in 1997, it pressured an Islamic-leaning prime minister, Necmettin Erbakan, to step down. Erbakan was the political mentor of current Prime Minister Recep Tayyip Erdogan.

A campus organization that officially formed Friday is asking the University to examine its contracts and purchases from companies that students say have ties to forced labor and trafficked children.
Free Project at GW, which includes six students, wants the University to become a ?global leader? against what they call slavery, pointing out that everything from food sold on campus to clothing sold in the bookstore could be produced in unsafe or unjust labor conditions.
The group has created an online petition, which gathered more than 400 signatures as of Sunday, demanding that the University becomes more conscious of potential instances of workers? rights violations. Melissa Arndorfer, who leads the group, said she hopes the petition, which went live in August, will hit 5,000 signatures by the end of December.
Lorena Martinez, a senior and a founding member of the group, said she thinks the University can realistically fulfill the group?s demands by taking ?baby steps.?
?It?s not going to happen overnight. We need to engage the entire University,? Martinez said, adding that she?s optimistic that GW will respond positively. Group members said they will officially present the petition to University President Steven Knapp after the petition gains more ground.
Arndorfer said the University should promote ethical purchases by learning more about its suppliers, looking at worker?s pay and fair trade certification to determine whether to take their business elsewhere.
?It?s the food I eat, the coffee I drink every day so I can wake up and go to class. My jeans, leather. It?s all these things,? Arndorfer said, citing her daily encounters with what she calls slavery.
The international affairs major said her group is not trying to ?antagonize? the University, but rather that it wants to set a new standard for the products and services offered at GW.
University spokeswoman Candace Smith said GW ?is committed to conducting its business affairs in a socially responsible manner.? She pointed out that GW is one of about 180 universities affiliated with the Worker Rights Consortium, which requires the University to verify that any official products that bear the school?s logo are checked to ensure ethical production.
Arndorfer said the group plans to raise awareness by handing out promotional material and using social media.
The catalyst for their campaign came from professor Michele Clark?s ?Human Trafficking? class in the Elliott School of International Affairs.
Clark, an expert in the issue, also helped Arndorfer create the petition.
?The cumulative value of this makes people really think about ethical purchasing, and it begins to trickle up [to the companies],? Clark said.
She said another examples of exploitation on campus are the adults she has seen supervising children a they sell candy around the Foggy Bottom metro stop. She said she believes it is an instance of child begging rings.
Clark said the University should call a human trafficking hotline and force the groups off campus to send a message that GW is not tolerant of human rights abuses.
